Inborn genetic diseases Uncertain:1
The c.482C>T (p.T161M) alteration is located in exon 4 (coding exon 4) of the CLN6 gene. This alteration results from a C to T substitution at nucleotide position 482, causing the threonine (T) at amino acid position 161 to be replaced by a methionine (M). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Ceroid lipofuscinosis, neuronal, 6A;C5561927:Ceroid lipofuscinosis, neuronal, 6B (Kufs type) Uncertain:1
CLN6 NM_017882 exon 4 p.Thr161Met (c.482C>T): This variant has not been reported in the literature but is present in 4/111698 European alleles in the Genome Aggregation Database (http://gnomad.broadinstitute.org/rs757734645). Evolutionary conservation and computational predictive tools for this variant are unclear. In summary, data on this variant is insufficient for disease classification. Therefore, the clinical significance of this variant is uncertain. -
Adult neuronal ceroid lipofuscinosis;C5551375:Ceroid lipofuscinosis, neuronal, 6A Uncertain:1
CLN6 NM_017882.2 exon 4 p.Thr161Met (c.482C>T): This variant has not been reported in the literature but is present in 4/111698 European alleles in the Genome Aggregation Database (http://gnomad.broadinstitute.org/rs757734645). Evolutionary conservation and computational predictive tools for this variant are unclear. In summary, data on this variant is insufficient for disease classification. Therefore, the clinical significance of this variant is uncertain. -
not provided Uncertain:1
A variant of uncertain significance has been identified in the CLN6 gene. The T161M variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. The T161M variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). The T161M variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In silico analysis predicts this variant is probably damaging to the protein structure/function. However, this substitution occurs at a position where amino acids with similar properties to Threonine are tolerated across species. Therefore, based on the currently available information, it is unclear whether this variant is a pathogenic variant or a rare benign variant. -
Neuronal ceroid lipofuscinosis Uncertain:1
This sequence change replaces threonine, which is neutral and polar, with methionine, which is neutral and non-polar, at codon 161 of the CLN6 protein (p.Thr161Met). This variant is present in population databases (rs757734645, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with CLN6-related conditions. ClinVar contains an entry for this variant (Variation ID: 452272). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
