The enzyme has both tyrosine hydroxylase and dopa oxidase catalytic activities, and requires copper for function. Mutations in this gene result in oculocutaneous albinism, and nonpathologic polymorphisms result in skin pigmentation variation. The TYR c.661G>A variant is predicted to result in the amino acid substitution p.Glu221Lys. This variant has been reported in the compound heterozygous state in individuals with oculocutaneous albinism (King et al. 2003. PubMed ID: 13680365; Gao et al. 2017. PubMed ID: 28451379). This variant has also been detected in the compound heterozygous state in an individual undergoing testing for hypopigmentation at PreventionGenetics (internal data). This variant is reported in 0.00088% of alleles in individuals of European (Non-Finnish) descent in gnomAD. Given the evidence, we interpret this variant as likely pathogenic. -
