The p.S234R variant (also known as c.700A>C), located in coding exon 5 of the SMAD4 gene, results from an A to C substitution at nucleotide position 700. The serine at codon 234 is replaced by arginine, an amino acid with dissimilar properties. This alteration was identified in an individual with aortopathy from China (Yang H et al. Sci Rep, 2016 09;6:33002). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
