Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with LAMB2-related disease. This variant is present in population databases (rs758825199, ExAC 0.005%). This sequence change replaces alanine with valine at codon 50 of the LAMB2 protein (p.Ala50Val). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and valine. -
