This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability. This variant is absent in the gnomAD Exomes. This variant causes a frameshift starting with codon Phenylalanine 1878, changes this amino acid to Leucine residue, and creates a premature Stop codon at position 39 of the new reading frame, denoted p.Phe1877LeufsTer39. It is submitted to ClinVar as Pathogenic/Likely pathogenic. This variant is predicted to cause loss of normal protein function through protein truncation. Loss of function variants have been previously reported to be disease causing. 5. However study on multiple affected individuals and functional studies on the pathogenicity of the variant is unavailable. For these reasons, this variant has been classified as Likely Pathogenic. -

This alteration, described as c.5631_5635delCTCGC+5639_5640insA, has been reported in two individuals from a cohort of Iranian ataxia-telangiectasia patients (Babaei M et al, Hum.
