This mutation has been associated with loss of heterozygosity in multiple tumors and aberrant splicing based on cDNA studies (Pelttari LM, Hum. Mol. Genet. 2011 Aug; 20(16):3278-88) and is recurrent in Finnish breast/ovarian cancer families, with haplotype analysis supporting a founder effect in this population (Nurmi A et al. Int J Cancer, 2019 11;145:2692-2700; Pelttari LM et al. Clin Genet, 2018 03;93:595-602). This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site; however, direct evidence is insufficient at this time (Ambry internal data). Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay.
