In the published literature, this variant and a truncating variant were reported in an individual affected with colorectal cancer (PMID: 19307944 (2009)). Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded conflicting predictions that this variant is deleterious or benign. Based on the available information, we are unable to determine the clinical significance of this variant. -

The p.S2710R variant (also known as c.8128A>C), located in coding exon 15 of the APC gene, results from an A to C substitution at nucleotide position 8128. The serine at codon 2710 is replaced by arginine, an amino acid with dissimilar properties. This alteration was identified in normal colon tissue in 1/117 colorectal cancer patients (Chen SP et al. Am J Clin Oncol, 2009 Apr;32:122-6). This amino acid position is not well conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Missense alterations in APC are not a common cause of disease (Spier I et al. Genet Med. 2024 Feb;26(2):100992). Based on the available evidence, the clinical significance of this variant remains unclear. -
