The p.S2556L variant (also known as c.7667C>T), located in coding exon 15 of the APC gene, results from a C to T substitution at nucleotide position 7667. The serine at codon 2556 is replaced by leucine, an amino acid with dissimilar properties. This variant has been reported in an individual affected with colorectal cancer (Yurgelun MB et al. J Clin Oncol, 2017 Apr;35:1086-1095). This variant was also detected in 1/113 Italian non-BRCA patients with a personal and/or familial history of breast cancer, ovarian cancer, and/or pancreatic cancer (Germani A et al. J Clin Med, 2020 Sep;9). This amino acid position is well conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This variant has been reported in an individual affected with colorectal cancer and an individual affected with breast cancer (PMID: 28135145, 32957588). This variant has been identified in 3/250832 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
