Variant summary: SLC26A4 c.445G>A (p.Gly149Arg) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251470 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.445G>A has been reported in the literature in individuals affected with clinical features of Pendred Syndrome (de Moraes_2013, Imizcoz_2023). These report(s) do not provide unequivocal conclusions about association of the variant with Pendred Syndrome. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in 55% of normal transport activity (de Moraes_2016).
