GHR (HGNC:4263): (growth hormone receptor) This gene encodes a member of the type I cytokine receptor family, which is a transmembrane receptor for growth hormone. Binding of growth hormone to the receptor leads to receptor dimerization and the activation of an intra- and intercellular signal transduction pathway leading to growth. Mutations in this gene have been associated with Laron syndrome, also known as the growth hormone insensitivity syndrome (GHIS), a disorder characterized by short stature. In humans and rabbits, but not rodents, growth hormone binding protein (GHBP) is generated by proteolytic cleavage of the extracellular ligand-binding domain from the mature growth hormone receptor protein. Variant summary: GHR c.497G>A (p.Gly166Glu) results in a non-conservative amino acid change located in the Fibronectin type III domain (IPR003961) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 250948 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.497G>A has been reported in the literature in individuals affected with short stature with non-growth-hormone deficiency and Congenital hypogonadotropic hypogonadism (Song_2006 and Chen_2020). These report(s) do not provide unequivocal conclusions about association of the variant with Growth Hormone Insensitivity. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 32502767, 32171629, 17274879, 36123965). ClinVar contains an entry for this variant (Variation ID: 225374). Based on the evidence outlined above, the variant was classified as uncertain significance. -
