The alanine at codon 420 is replaced by threonine, an amino acid with similar properties. This variant was identified in a patient with cystic-papillary serous carcinoma of the ovary at age 28, slight perioral hyperpigmentation and facial freckling; however, she had no reported history of gastrointestinal problems, Peutz-Jeghers syndrome or cancer in her family (Heinritz W et al., Onkologie 2008 Nov; 31(11):625-8). This amino acid position is not well conserved in available vertebrate species, and threonine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
