It shares with other type I receptors a high degree of similarity in serine-threonine kinase subdomains, a glycine- and serine-rich region (called the GS domain) preceding the kinase domain, and a short C-terminal tail. The encoded protein, sometimes termed ALK1, shares similar domain structures with other closely related ALK or activin receptor-like kinase proteins that form a subfamily of receptor serine/threonine kinases. This sequence change replaces leucine, which is neutral and non-polar, with glutamine, which is neutral and polar, at codon 123 of the ACVRL1 protein (p.Leu123Gln). This variant is present in population databases (rs762756031, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with ACVRL1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
