Myosin is a hexameric protein composed of a pair of myosin heavy chains (MYH) and two pairs of nonidentical light chains. This gene is a member of the MYH family and encodes a protein with an IQ domain and a myosin head-like domain. Mutations in this gene have been associated with two congenital contracture (arthrogryposis) syndromes, Freeman-Sheldon syndrome and Sheldon-Hall syndrome. [provided by RefSeq, Jul 2008]

The variant was not present in any of the unaffected family members and was noted to segregate with disease (Alvarado et al. 2011). Control data are unavailable for this variant which is not found in the 1000 Genomes Project, the Exome Sequencing Project, or the Exome Aggregation Consortium. The evidence for this variant is limited to a single family. The p.Phe437Ile variant is therefore classified as a variant of unknown significance, but suspicious for pathogenicity for MYH3-related disorders. This variant was observed by ICSL as part of a predisposition screen in an ostensibly healthy population. - |
