The previously defined group FANCH is the same as FANCA. Fanconi anemia is a genetically heterogeneous recessive disorder characterized by cytogenetic instability, hypersensitivity to DNA crosslinking agents, increased chromosomal breakage, and defective DNA repair. The members of the Fanconi anemia complementation group do not share sequence similarity; they are related by their assembly into a common nuclear protein complex. This frequency is not significantly higher than estimated for a pathogenic variant in FANCE causing Fanconi Anemia (6e-05 vs 0.00048), allowing no conclusion about variant significance. c.598C>T has been reported in the literature in individuals affected with Colon cancer, Breast cancer and other unspecified childhood cancer, without strong evidence for causality (example, Esteban-Jurado_2016, Sylvester_2021). These report(s) do not provide unequivocal conclusions about association of the variant with Fanconi Anemia. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 27165003, 34687117).
