Variant summary: ATM c.3402+16A>G alters a non-conserved nucleotide located at a position not widely known to affect splicing. Consensus agreement among computation tools predict no significant impact on normal splicing (TrAP). However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 3.2e-05 in 250100 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3402+16A>G has been reported in the literature in individuals affected with colorectal cancer, without strong evidence for causality (Erdem_2020). These report(s) do not provide unequivocal conclusions about association of the variant with Ataxia-Telangiectasia.
