The p.N568K variant (also known as c.1704T>G), located in coding exon 11 of the BRIP1 gene, results from a T to G substitution at nucleotide position 1704. The asparagine at codon 568 is replaced by lysine, an amino acid with similar properties. One study detected this alteration in 2/13213 breast cancer cases and 0/5242 population controls (Easton DF et al. J. Med. Genet., 2016 05;53:298-309). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
