The c.1888dupA pathogenic mutation, located in coding exon 12 of the BRIP1 gene, results from a duplication of A at nucleotide position 1888, causing a translational frameshift with a predicted alternate stop codon (p.T630Nfs*9). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
This variant inserts 1 nucleotide in exon 13 of the BRIP1 gene, creating a frameshift and premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. This variant has been reported in an individuals affected with breast cancer and ovarian cancer (PMID: 26921362, 33471991, 36315097, 36765687). This variant has also been identified in 1/251444 chromosomes in the general population by the Genome Aggregation Database (gnomAD).
