The encoded protein is believed to be an integral membrane protein involved in small molecule transport, specifically tyrosine, which is a precursor to melanin synthesis. It is involved in mammalian pigmentation, where it may control skin color variation and act as a determinant of brown or blue eye color. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251488 control chromosomes (gnomAD). c.1427A>G has been reported in the literature in individuals affected with Oculocutaneous Albinism (Preising_2007, Mauri_2016, Lasseaux_2018). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 29345414, 27734839, 17960121). ClinVar contains an entry for this variant (Variation ID: 211767). Based on the evidence outlined above, the variant was classified as likely pathogenic. -
