The p.V405A variant (also known as c.1214T>C), located in coding exon 10 of the CEP57 gene, results from a T to C substitution at nucleotide position 1214. The valine at codon 405 is replaced by alanine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -

This sequence change replaces valine, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 405 of the CEP57 protein (p.Val405Ala).
