Calsequestrin is localized to the sarcoplasmic reticulum in cardiac and slow skeletal muscle cells. The protein is a calcium binding protein that stores calcium for muscle function. Mutations in this gene cause stress-induced polymorphic ventricular tachycardia, also referred to as catecholaminergic polymorphic ventricular tachycardia 2 (CPVT2), a disease characterized by bidirectional ventricular tachycardia that may lead to cardiac arrest. Hum Mutat, 2011 Sep;32:995-9). This variant has also been detected in a case of sudden infant death and an additional CVPT cohort; however, details were limited (Tester DJ et al. J Am Coll Cardiol, 2018 Mar;71:1217-1227). (Ng K et al. Circulation, 2020 Sep;142:932-947). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
