This sequence change replaces aspartic acid, which is acidic and polar, with tyrosine, which is neutral and polar, at codon 157 of the STN1 protein (p.Asp157Tyr). This variant is present in population databases (rs765462548, gnomAD 0.003%). This missense change has been observed in individual(s) with Coats plus syndrome and/or primary immunodeficiency (PMID: 27432940, 32135276). ClinVar contains an entry for this variant (Variation ID: 375592). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt STN1 protein function with a positive predictive value of 80%.
