This sequence change replaces lysine, which is basic and polar, with arginine, which is basic and polar, at codon 194 of the NECAP1 protein (p.Lys194Arg). This variant is present in population databases (rs765855460, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with NECAP1-related conditions. ClinVar contains an entry for this variant (Variation ID: 475007). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
