This sequence change replaces lysine with arginine at codon 377 of the MEN1 protein (p.Lys377Arg). The lysine residue is highly conserved and there is a small physicochemical difference between lysine and arginine. This variant has not been reported in the literature in individuals with MEN1-related disease. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. -

The p.K377R variant (also known as c.1130A>G), located in coding exon 7 of the MEN1 gene, results from an A to G substitution at nucleotide position 1130. The lysine at codon 377 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
