This variant removes 4 nucleotides at the +4 to +7 positions in the intron 13 splice donor site of the BRIP1 gene. Computational splicing tools predict that this variant may have a significant impact on RNA splicing. To our knowledge, RNA analysis studies have not been reported for this variant nor has this variant been reported in individuals affected with hereditary cancer in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The c.1935+4_1935+7delAGTT intronic variant, located in intron 12 of the BRIP1 gene, results from a deletion of 4 nucleotides within intron 12 of the BRIP1 gene. These nucleotide positions are generally well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and may result in the creation or strengthening of a novel splice donor site; however, direct evidence is insufficient at this time (Ambry internal data). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
