This variant removes 4 nucleotides at the +4 to +7 positions in the intron 13 splice donor site of the BRIP1 gene. Computational splicing tools predict that this variant may have a significant impact on RNA splicing. To our knowledge, RNA analysis studies have not been reported for this variant nor has this variant been reported in individuals affected with hereditary cancer in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The c.1935+4_1935+7delAGTT intronic variant begins 4 nucleotides after coding exon 12 in the BRIP1 gene. This variant results from a deletion of 4 nucleotides at positions c.1935+4 to c.1935+7. These nucleotide positions are well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and may result in the creation or strengthening of a novel splice donor site; however, direct evidence is insufficient at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear. -
