Disruption of the orthologous mouse gene shows that it plays a role in auditory and vestibular functions. It is involved in fibrillar network organization, the anchoring of otoconial membranes and cupulae to the neuroepithelia, and likely in sound stimulation resistance. Mutations in this gene cause autosomal recessive nonsyndromic deafness, type 18B. Alternative splicing of this gene results in multiple transcript variants. [provided by RefSeq, May 2014]

This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 2691 of the OTOG protein (p.Arg2691His). This variant is present in population databases (rs766229978, gnomAD 0.07%). This variant has not been reported in the literature in individuals affected with OTOG-related conditions. ClinVar contains an entry for this variant (Variation ID: 227805). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Possibly Damaging". The histidine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
