The threonine at codon 3033 is replaced by proline, an amino acid with highly similar properties. This alteration has been detected in 2/2575 unselected patients with breast cancer and was not detected in 2809 healthy control individuals from a Malaysian cohort (Wen WX et al. J. Med. Genet., 2018 02;55:97-103). This alteration has been previously identified in a cohort of 533 Malaysian breast cancer cases with <10% a priori risk for a BRCA1/2 mutation (Kang PC et al. Breast Cancer Res. Treat., 2014 Apr;144:635-42). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |

Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 3.9e-06 in 255078 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.9097A>C has been reported in the literature in individuals affected with Breast Cancer (Kang_2014, Wen_2018). These reports do not provide unequivocal conclusions about association of the variant with Hereditary Breast and Ovarian Cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. - |
