The p.E235G variant (also known as c.704A>G), located in coding exon 3 of the GALNT12 gene, results from an A to G substitution at nucleotide position 704. The glutamic acid at codon 235 is replaced by glycine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces glutamic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 235 of the GALNT12 protein (p.Glu235Gly). This variant is present in population databases (rs767946700, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with GALNT12-related conditions. ClinVar contains an entry for this variant (Variation ID: 241515).
