The arginine at codon 777 is replaced by cysteine, an amino acid with highly dissimilar properties. In one study, the p.R777C alteration was detected in 1/101,759 breast cancer cases and 0/15,587 ovarian cancer cases, and in an inter-strand cross link damage survival assay, the p.R777C alteration was found to be functionally abnormal (Moyer CL et al. Cancer Res. 2020 Feb;80:857-867). This alteration was identified in an individual diagnosed with Fanconi anemia (George M et al. Hum Mutat, 2021 Dec;42:1648-1665). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
