Hypercholesterolemia, familial, 1 Uncertain:2
This missense variant (also known as p.Glu629Lys in the mature protein) replaces glutamic acid with lysine at codon 650 of the LDLR protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual with elevated cholesterol levels and a family history of heart disease (PMID: 20236128). This variant has been identified in 13/251472 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

Familial hypercholesterolemia Uncertain:1
This missense variant replacesglutamic acid with lysine at codon 650 of the LDLR protein. This variant is also known as p.Glu629Lys in the mature protein. Computational prediction tools indicate that this variant's impact on protein structure and function is inconclusive. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in two individual affected with familial hypercholesterolemia (PMID: 20236128, 23375686). This variant has been identified in 13/251472 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
