Variant summary: IGHMBP2 c.1274G>A (p.Arg425His) results in a non-conservative amino acid change located in the Helicase superfamily 1/2, ATP-binding domain (IPR014001) of the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 2.4e-05 in 248022 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1274G>A has been observed in individual(s) affected with Charcot-Marie-Tooth disease axonal type 2S, however the variant was found in cis with other variants with unknown classifications (Quaio_2020). These report(s) do not provide unequivocal conclusions about association of the variant with Charcot-Marie-Tooth disease axonal type 2S. A different variant affecting the same codon has been classified as pathogenic by our lab (c.1273C>T, p.Arg425Cys), supporting the critical relevance of codon 425 to IGHMBP2 protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
