Hereditary cancer-predisposing syndrome Uncertain:2
The p.S703Y variant (also known as c.2108C>A), located in coding exon 13 of the RAD50 gene, results from a C to A substitution at nucleotide position 2108. The serine at codon 703 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This sequence change replaces serine, which is neutral and polar, with tyrosine, which is neutral and polar, at codon 703 of the RAD50 protein (p.Ser703Tyr). This variant is present in population databases (rs769143998, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with RAD50-related conditions. ClinVar contains an entry for this variant (Variation ID: 480392). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt RAD50 protein function with a positive predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
RAD50-related disorder Uncertain:1
The RAD50 c.2108C>A variant is predicted to result in the amino acid substitution p.Ser703Tyr. This variant has been reported as a rare germline variant in an individual with cancer from the TCGA discovery cohort (Supplementary Data 12, Lu C et al. 2015. PubMed ID: 26689913). This variant is reported in 0.012% of alleles in individuals of African descent in gnomAD (http://gnomad.broadinstitute.org/variant/5-131931403-C-A). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
