This variant has not been reported in the literature in individuals affected with POLE-related conditions. ClinVar contains an entry for this variant (Variation ID: 566551). RNA analysis performed to evaluate the impact of this premature translational stop signal on mRNA splicing indicates it does not significantly alter splicing (internal data). The p.R249* variant (also known as c.745C>T), located in coding exon 8 of the POLE gene, results from a C to T substitution at nucleotide position 745. This changes the amino acid from an arginine to a stop codon within coding exon 8. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This region is highly conserved in available vertebrate species. As neither this specific alteration nor loss of function as a mechanism of pathogenicity have been well-described in the POLE gene, the clinical significance of this variant remains unknown (ACMG Standards and guidelines for the interpretation of sequence variants.
