This sequence change replaces alanine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 1059 of the TRAPPC9 protein (p.Ala1059Val). This variant is present in population databases (rs769398594, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with TRAPPC9-related conditions. The p.A1059V variant (also known as c.3176C>T), located in coding exon 20 of the TRAPPC9 gene, results from a C to T substitution at nucleotide position 3176. The alanine at codon 1059 is replaced by valine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear. -
