This missense variant replaces isoleucine with valine at codon 233 of the BRIP1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). Splice site prediction tools suggest that this variant may not impact RNA splicing. To our knowledge, functional studies have not been performed for this variant. This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 1/251144 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The p.I233V variant (also known as c.697A>G), located in coding exon 6 of the BRIP1 gene, results from an A to G substitution at nucleotide position 697. The isoleucine at codon 233 is replaced by valine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. Variant summary: BRIP1 c.697A>G (p.Ile233Val) results in a conservative amino acid change located in the Helicase superfamily 1/2, ATP-binding domain, DinG/Rad3-type of the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 245854 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.697A>G in individuals affected with Hereditary Breast and Ovarian Cancer and no experimental evidence demonstrating its impact on protein function have been reported. One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation, and classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -
