Variant summary: EFHC1 c.547G>A (p.Val183Ile) results in a conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 2.8e-05 in 250982 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.547G>A in individuals affected with Juvenile Myoclonic Epilepsy and no experimental evidence demonstrating its impact on protein function have been reported. The valine residue is weakly conserved and there is a small physicochemical difference between valine and isoleucine. This variant is present in population databases (rs769591944, ExAC 0.02%). This variant has not been reported in the literature in individuals with EFHC1-related disease. ClinVar contains an entry for this variant (Variation ID: 205378). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The isoleucine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function.
