The p.V17340I variant (also known as c.52018G>A), located in coding exon 153 of the TTN gene, results from a G to A substitution at nucleotide position 52018. The valine at codon 17340 is replaced by isoleucine, an amino acid with highly similar properties. This alteration (referred to as p.V23837I, c.71508G>A) has been reported as a secondary cardiac variant in an exome cohort (Ng D et al. Circ Cardiovasc Genet, 2013 Aug;6:337-46). This amino acid position is well conserved in available vertebrate species; however, isoleucine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
