The encoded protein binds actin and interacts with thin filaments and Z-line associated proteins in striated muscle. This protein may be involved in cardiac myofibril assembly. A shorter isoform of this protein termed LIM nebulette is expressed in non-muscle cells and may function as a component of focal adhesion complexes. The p.P671L variant (also known as c.2012C>T), located in coding exon 20 of the NEBL gene, results from a C to T substitution at nucleotide position 2012. The proline at codon 671 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 671 of the NEBL protein (p.Pro671Leu). This variant is present in population databases (rs770107953, gnomAD 0.03%). This variant has not been reported in the literature in individuals affected with NEBL-related conditions.
