MEN1-related disorder Uncertain:1
The MEN1 c.769G>T variant is predicted to result in the amino acid substitution p.Asp257Tyr. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.012% of alleles in individuals of African descent in gnomAD (http://gnomad.broadinstitute.org/variant/11-64575053-C-A), which may be too common to be an undocumented primary cause of disease. Although we suspect that this variant may be benign, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
Hereditary cancer-predisposing syndrome Uncertain:1
The p.D252Y variant (also known as c.754G>T), located in coding exon 3 of the MEN1 gene, results from a G to T substitution at nucleotide position 754. The aspartic acid at codon 252 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
Multiple endocrine neoplasia, type 1 Benign:1
