The p.T2471A variant (also known as c.7411A>G), located in coding exon 13 of the BRCA2 gene, results from an A to G substitution at nucleotide position 7411. The threonine at codon 2471 is replaced by alanine, an amino acid with similar properties. In one study, this alteration was observed in 1/12490 male controls and not detected in 7051 unselected female breast cancer patients, 53 male breast cancer patients or 11241 female controls of Japanese ancestry (Momozawa Y et al. Nat Commun, 2018 10;9:4083). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces threonine with alanine at codon 2471 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 2/251000 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

The variant allele was found at a frequency of 2.2e-05 in 277658 control chromosomes (genomAD and publication data). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.7411A>G has been reported in the literature in individuals affected with Hereditary Breast And Ovarian Cancer Syndrome or prostate cancer (Kim_2020, Park_2020, Park_2021, So_2022). These reports do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 31837001, 31907386, 33875706, 30287823, 34063308, 32812259, 35534218). Three submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. All submitters classified the variant as uncertain significance.
