This variant is present in population databases (rs771127702, ExAC 0.07%), and has an allele count higher than expected for a pathogenic variant (PMID: 28166811). This sequence change replaces cysteine with arginine at codon 1114 of the SCN10A protein (p.Cys1114Arg). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and arginine. -
