This sequence change replaces arginine with histidine at codon 173 of the EPCAM protein (p.Arg173His). The arginine residue is highly conserved and there is a small physicochemical difference between arginine and histidine. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C25"). This variant is present in population databases (ExAC <0.01%) but has not been reported in the literature. ClinVar contains an entry for this variant (Variation ID: 216559). In summary, this is a rare missense change with uncertain impact on protein function. Although there is no indication that this variant causes disease, the evidence is insufficient at this time to prove that conclusively.
