The p.M182T variant (also known as c.545T>C), located in coding exon 1 of the HOXB13 gene, results from a T to C substitution at nucleotide position 545. The methionine at codon 182 is replaced by threonine, an amino acid with similar properties. One study found that this alteration was predicted to be deleterious by multiple in silico prediction models (Chandrasekaran G et al. Sci Rep, 2017 Mar;7:43830). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
