The variant has been reported to be in trans with a pathogenic variant as either compound heterozygous or homozygous in at least 2 similarly affected unrelated individuals(PMID: 30117111, PM3_S). The variant was co-segregated with Myasthenic syndrome, congenital, 13, with tubular aggregates in multiple affected family members (PMID: 30117111, PP1_P). In silico tool predictions suggest damaging effect of the variant on gene or gene product (3CNET: 0.785, PP3_P). A missense variant is a common mechanism associated with Myasthenic syndrome (PP2_P). It is observed at an extremely low frequency in the gnomAD v2.1.1 dataset (total allele frequency: 0.000016, PM2_M). Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2.5e-05 in 279668 control chromosomes. c.739C>T has been reported in the literature in individuals affected with Congenital Disorder Of Glycosylation Type 1J (Jones_2013, Ng_2019, Cao_2021). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
