This gene encodes a muscle-specific member of the calpain large subunit family that specifically binds to titin. Mutations in this gene are associated with limb-girdle muscular dystrophies type 2A. This missense change has been observed in individuals with autosomal recessive limb-girdle muscular dystrophy (PMID: 15221789, 16141003, 18055493, 20517216, 23821418, 26484845; Invitae). It has also been observed to segregate with disease in related individuals. This variant has been reported in individual(s) with autosomal dominant limb-girdle muscular dystrophy (PMID: 20517216); however, the role of the variant in this condition is currently unclear. ClinVar contains an entry for this variant (Variation ID: 288404). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt CAPN3 protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change affects CAPN3 function (PMID: 11371436). For these reasons, this variant has been classified as Pathogenic. -
