This sequence change replaces glycine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 134 of the GLB1 protein (p.Gly134Val). This variant is present in population databases (rs773562141, gnomAD 0.0009%). This missense change has been observed in individual(s) with GM1-gangliosidosis and/or mucopolysaccharidosis type IVB (PMID: 17309651, 33558080). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 556153). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt GLB1 protein function with a positive predictive value of 95%. This variant disrupts the p.Gly134 amino acid residue in GLB1. Other variant(s) that disrupt this residue have been observed in individuals with GLB1-related conditions (PMID: 25936995), which suggests that this may be a clinically significant amino acid residue.
