The c.3634_3639delAATGAA variant (also known as p.N1212_E1213del) is located in coding exon 10 of the BRCA2 gene. This variant results from an in-frame AATGAA deletion at nucleotide positions 3634 to 3639. This results in the in-frame deletion of 2 amino acids (NE) at codons 1212 and 1213. These amino acid positions are not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear. -
