The p.R2090W variant (also known as c.6268C>T), located in coding exon 38 of the DNAH5 gene, results from a C to T substitution at nucleotide position 6268. The arginine at codon 2090 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

Variant summary: DNAH5 c.6268C>T (p.Arg2090Trp) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251174 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.6268C>T has been reported in the literature in at-least one individual affected with Primary ciliary dyskinesia (Noethe-Menchen_2019). These report(s) do not provide unequivocal conclusions about association of the variant with Primary ciliary dyskinesia 3. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 31638833). ClinVar contains an entry for this variant (Variation ID: 566691).
