Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with KCNH2-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces isoleucine with methionine at codon 655 of the KCNH2 protein (p.Ile655Met). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and methionine. -

The p.I655M variant (also known as c.1965C>G), located in coding exon 8 of the KCNH2 gene, results from a C to G substitution at nucleotide position 1965. The isoleucine at codon 655 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
