Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 1.6e-05 in 251258 control chromosomes (gnomAD). c.2050G>A has been found in individuals affected with Familial Hypercholesterolemia (e.g. Khoo_2000, Reijman_2023, internal data). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
