The proline at codon 441 is replaced by leucine, an amino acid with similar properties. This alteration was observed in 1/431 individuals with microsatellite stable colorectal cancer (Stenzinger A et al. Cancer Med. 2014 Dec;3:1527-38). It was also reported in a 64 year old male with Lynch-like syndrome (LLS) in a cohort of 81 patients with LLS and 47 patients with Lynch syndrome who all had mismatch repair deficient colorectal cancer (Xu Y et al. Front Genet, 2020 Aug;11:991)This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
